His early career was diverse, seeing him work as a radio announcer in Montreal and appearing as a live-action performer on programs like The Smothers Brothers Comedy Hour and The Sonny & Cher Comedy Hour. However, it was his transition into the world of voice acting that would eventually make him a household name.

In 1984, Cullen auditioned for a new project called The Transformers. At the time, the premise was simple but unusual: he was told the character was a hero, but he was also a truck. To find the right tone for the leader of the Autobots, Cullen looked toward his older brother, Larry, a decorated U.S. Marine who had served in Vietnam. The contrast between the booming authority of a robotic leader and the soft, depressive tones of Eeyore highlighted his immense range as a performer. He often shared how much he enjoyed the reactions of young children when he would drop into Eeyore’s voice to say, "Hello. Thanks for noticing me".

His vocal talents extended into the realm of science fiction and horror as well. Cullen was the man behind the terrifying, clicking vocalizations of the creature in the 1987 film Predator. He famously insisted on seeing the creature’s design before creating the sound, noting that the "ugly, ugly face" reminded him of an upside-down horseshoe crab dying in the sun. He also voiced the 1976 remake of King Kong and the sinister K.A.R.R. on Knight Rider.

Beyond the recording booth, Cullen was deeply involved in charitable and educational efforts. He worked with NASA on the Optimus Prime Spinoff Award, which encourages students to pursue interests in space, science, and technology. His commitment to inspiring the next generation was a hallmark of his later years.

He remained humble about his success, once stating that being a part of the Transformers phenomenon was "historical" and something he was deeply grateful for.
